A friend of mine just sent me this link:

He is working remotely for a Japan company and his employer just realized that one of the backers of his kickstarter project is the guy from the breaking news today.

"Mark Karpeles   http://www.tibanne.com/   PHP can do stuff!"

The interesting part is that today he changed the referral site from http://www.mtgox.com/ to http://www.tibanne.com/

He said he was a "very generous donor"

"It looks like he was an active backer on KickStarter, backing over 100 projects with the most recent ending less than a month ago. Many of the projects he backed were Open Source software or hardware."

"every project he backed is a project I would back. He has extremely good taste"

"I've had no contact with him other than the kickstarter survey though. He has not repsponed to any of may messages through KS"

Reading the chatlog i came up with a theory that looks increasingly probable to me.
(until i read prof7bit's explanation which came up while typing this and can be even closer to the truth but i can't comment on the technical side)

Someone's gone missing with the second/third key.Either kidnapped or gone rogue,probably blackmailing
"technically speaking it's not "lost" just yet, just temporarily unavailable"

This ties in with the "security concerns" the other guys will need his key also and "third parties involved"
Looks like he sees the coins not spent but he can't control them for some reason.

That can also explain how the document has some real figures and maybe things they were discussing among them if that is what he is admitting with "more or less" but "this document was not produced by MtGox".

Mark has always appeared to me like a shy nerd,currently in deep trouble.

Other variation on this is if he is in regulatory trouble (maybe something similar to Charley Shrem situation) which is preventing him from accessing the deposit box in which the keys are stored.

Well i am looking for more indepth answers as well but here is my take:

Back when gox was suffering from stuck transaction someone made a graph to track the transactions broadcasted by the gox api.Those of them that were not included in the blockchain showed as "stuck" accumulating to 70k at the peak.It was believed that this was happening because some of the inputs of those transactions were from newly mined coins which can't be spent before 100 confirmations.Then when tx malleability came to the spotlight that was the most probable explanation-that these inputs were already spent with other txID.

Now for the first time since gox closed a big 20k transaction was broadcasted by their api.It showed as a spike in the stuck transaction graph till it was included in the blockchain.

Today there is a 200k address that is being followed by the community,previously linked to gox wallets which is spitted 60:40% at every new mined block.

This post by Gregory Maxwell makes a link between the 20k transaction from gox api and 200k coins that are moving today.
